Variation of HbA1c with Hemoglobin level: A comparative study between High Performance Liquid Chromatography and Immunochromatography Analyzer

Introduction: The measurement of glycated hemoglobin (HbA1c) is an established procedure in evaluating long-term glycemic control diabetic patients. There are many factors like temperature, pH, lifespan protein, substrate concentrations and levels that influence HbA1c estimation by any methods. aim the study to estimate High Performance Liquid Chromatography (HPLC) immunochromatography analyzer find both Methods: This was a descriptive, cross-sectional carried out department Biochemistry Shree Birendra Hospital, Chhauni, Kathmandu over period two months from May 2021 June 2021. We selected patients with type 2 diabetes mellitus. measured EDTA blood samples Bio-Rad D10 (HPLC method) Nycocard (Immunochromatography whereas estimated using Horiba Penta XLR fully automated hematology analyzer. Results: In our 100 known cases mellitus, mean level 14 ± 2.09 g/dL. value obtained HPLC were 7 1.95 % 7.39 2.21% respectively. found there strong correlation values method p = 0.001, r 0.6. However, we no such 0.6, 0.08. Conclusion: positive however variation seen method. suggests estimating methods may show decreased increased levels.
